NTyP-100 is a novel nanocarrier-based therapeutic candidate developed by Aqualung Therapeutics for the treatment of acute lung injury (ALI), ventilator-induced lung injury (VILI), and other severe inflammatory and fibrotic diseases. The nanocarrier is designed to restore vascular barrier integrity and reduce vascular permeability. It delivers a dual-action therapeutic cargo consisting of conjugated tysiponate (TySIP), a sphingosine-1-phosphate (S1P) analogue that targets S1P receptors (primarily S1PR1) to enhance endothelial barrier function, and an encargoed peptide inhibitor (PIK) of non-muscle myosin light chain kinase (nmMLCK) to block myosin light chain hyperphosphorylation and prevent paracellular gap formation. In preclinical rodent models of lipopolysaccharide (LPS)-induced acute lung injury and ventilator-induced lung injury, intravenous administration of NTyP-100 significantly reduced lung inflammation, vascular leakage, and inflammatory cell infiltration.
