NUC-3373 + bevacizumab is an investigational combination therapy for cancer, specifically metastatic colorectal cancer. NUC-3373 is a novel small molecule derived from the nucleoside analog 5-fluorouracil, designed to overcome resistance mechanisms and improve efficacy compared to standard fluoropyrimidines. It acts as an antimetabolite, interfering with DNA synthesis in rapidly dividing tumor cells. Bevacizumab is a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis required for tumor growth and metastasis. The combination aims to enhance anti-tumor activity by both direct cytotoxic effects on cancer cells (via NUC-3373) and inhibition of tumor blood vessel formation (via bevacizumab). This regimen has been evaluated in clinical trials for second-line treatment of metastatic colorectal cancer, often alongside other agents such as leucovorin, irinotecan or oxaliplatin[5][7].
