NUC B1000 is an expressed interfering RNA (eiRNA)-based therapeutic developed by Nucleonics, Inc. for the treatment of chronic hepatitis B virus (HBV) infection. The drug consists of a plasmid DNA construct designed to produce four distinct short interfering RNA (siRNA) molecules, formulated within a proprietary cationic-lipid delivery system. Each of the four siRNAs targets a different conserved sequence of the HBV genome, collectively designed to degrade all viral RNA species produced within infected cells. This multi-target approach aims to prevent viral escape and effectively inhibit HBV replication and protein expression. NUC B1000 entered Phase I clinical testing in 2008 to evaluate safety and tolerability in patients with chronic HBV, but development was subsequently discontinued following the closure of Nucleonics.
