NUFIRI

NUFIRI is an investigational combination chemotherapy regimen developed by NuCana plc for the treatment of advanced or metastatic colorectal cancer (CRC). The regimen consists of NUC-3373 (fosifloxuridine nafalbenamide), leucovorin, and irinotecan. NUC-3373 is a ProTide transformation of 5-fluorodeoxyuridine monophosphate (FUDR-MP) designed to bypass key resistance mechanisms associated with 5-fluorouracil (5-FU) by delivering the active metabolite directly into cells and inhibiting thymidylate synthase more effectively. Irinotecan, a topoisomerase I inhibitor, is included to provide synergistic DNA-damaging effects. The regimen was evaluated in the Phase II NuTide:323 study as a potential replacement for the standard-of-care FOLFIRI regimen. However, development for second-line colorectal cancer was discontinued in 2024 after an interim analysis suggested the regimen was unlikely to demonstrate superior progression-free survival compared to FOLFIRI plus bevacizumab.
