Drug intelligence / Profile preview
Nutropin Depot is a long-acting, injectable formulation of recombinant human growth hormone (somatropin) that utilizes Alkermes' ProLease injectable extended-release technology. It consists of somatropin encapsulated in biodegradable polylactide-co-glycolide (PLG) microspheres, designed to provide a sustained release of the hormone over several weeks. This formulation was developed through a collaboration between Genentech and Alkermes to reduce the injection frequency for patients with growth hormone deficiency (GHD) from daily to once or twice monthly. Somatropin acts as an agonist at the growth hormone receptor (GHR), stimulating the production of insulin-like growth factor 1 (IGF-1) and promoting linear growth, protein synthesis, and metabolic regulation. Although it was approved by the FDA in 1999, Genentech voluntarily withdrew Nutropin Depot from the market in 2004 due to the significant manufacturing resources required to produce the drug.
