**NUV-1156** is a preclinical, chimeric small-molecule drug-drug conjugate developed by Nuvation Bio for androgen receptor-positive cancers, principally prostate cancer. It covalently combines an enzalutamide-derived androgen receptor-binding moiety with an olaparib-derived poly(ADP-ribose) polymerase inhibitor warhead. The design aims to concentrate PARP-inhibitory cytotoxicity in androgen receptor-expressing tumor cells while reducing exposure in low-androgen-receptor normal tissues such as gastrointestinal tract and bone marrow. It showed preclinical activity in enzalutamide-resistant prostate cancer models and was also evaluated in triple-negative breast cancer cell lines.
