Overview

NUV-569 is a small molecule, selective inhibitor of **Wee1 kinase** being developed by Nuvation Bio. Wee1 is a serine/threonine-protein kinase that regulates the G2-M cell cycle checkpoint, allowing cells time to repair DNA damage before mitosis. Inhibition of Wee1 can force tumor cells with unrepaired DNA damage to enter mitosis, leading to catastrophic DNA damage and cell death. NUV-569 was designed to improve kinase selectivity and avoid off-target activity, aiming to increase therapeutic window and tolerability compared to earlier Wee1 inhibitors. Its intended clinical role was to synergize with DNA damaging therapies (e.g., chemotherapy, radiation) to enhance anti-tumor activity, especially in solid tumors such as pancreatic cancer. Nuvation Bio deprioritized the development of NUV-569 in 2023, and its development is currently discontinued[1][3][5][7][9][12][13].
