Overview

NV1066 is a replication-competent, attenuated oncolytic herpes simplex virus type 1 (HSV-1) engineered for the treatment of various solid tumors. It is characterized by deletions in the γ134.5 (ICP34.5) and ICP47 genes, which restrict viral replication to neoplastic cells and enhance the presentation of viral and tumor antigens. NV1066 is further modified to express the enhanced green fluorescent protein (eGFP) for tracking viral distribution and the viral thymidine kinase (TK) gene. The expression of TK allows for a synergistic "bystander effect" when administered alongside the prodrug ganciclovir, as the activated drug can spread to neighboring uninfected cells via gap junctions or tunneling nanotubes, inducing apoptosis in non-infected recipient cells.
