Overview

NV651 is a second-generation, sanglifehrin-based small molecule inhibitor of cyclophilins, developed by **Abliva** (formerly **NeuroVive Pharmaceutical**). It specifically targets the peptidyl-prolyl isomerase (PPIase) activity of multiple cyclophilin isoforms, including **Cyclophilin A (PPIA)**, **Cyclophilin B (PPIB)**, and **Cyclophilin D (PPIF)**. These proteins are often overexpressed in various malignancies, particularly **hepatocellular carcinoma (HCC)**, where they contribute to tumor cell proliferation, survival, and resistance to chemotherapy. NV651 exerts its anti-cancer effects by inducing G2/M phase cell cycle arrest and triggering intrinsic apoptosis. Preclinical studies have demonstrated that NV651 can work synergistically with DNA-damaging agents like **cisplatin**, potentially by downregulating DNA repair pathways and modulating the expression of genes involved in the cancer cell cycle.
