Overview

NVG-222 is a next-generation, half-life extended, bispecific T-cell engager developed by NovalGen. It is designed to target ROR1, a protein highly expressed on certain cancer cells, and CD3, a protein found on T cells. This dual targeting mechanism aims to bring T cells into close proximity with cancer cells, thereby activating the T cells to kill the cancer cells. A key differentiating feature of NVG-222 is NovalGen's proprietary AutoRegulation (AR) technology. This technology enables the drug to self-inactivate in response to excessive T-cell activation, which helps to mitigate mechanism-of-action-related toxicities, such as cytokine release syndrome, and improve the overall safety and durability of the immunotherapy. NVG-222 is being developed for the treatment of ROR1-positive hematological malignancies and solid tumors. It builds upon the clinical success of NVG-111, a precursor molecule that shares the same ROR1- and CD3-binding domains but lacks the AR functionality.
