Overview

NVG82 is a novel, engineered adeno-associated virus (AAV) capsid developed by REGENXBIO for ocular gene therapy. Identified through peptide insertion-based library selection from an AAV8-derived library, NVG82 is optimized for delivery via the suprachoroidal space (SCS). It demonstrates significantly enhanced transduction efficiency in the retinal pigment epithelium (RPE) and outer retinal layers, including photoreceptors, compared to the parental AAV8 capsid. Mechanistically, NVG82 exhibits a unique binding profile, interacting with heparin and protein receptors such as the adeno-associated virus receptor (AAVR) and carboxypeptidase D (CPD/AAVR2), which facilitates its ability to cross ocular tissue barriers and achieve robust transgene expression at lower doses.
