NVP-ADW742 is a small molecule kinase inhibitor that selectively targets the insulin-like growth factor 1 receptor (IGF-1R). It has demonstrated potent inhibition of IGF-1R kinase activity (IC50 of 0.17 μM) and is more than 16-fold more selective for IGF-1R compared to the insulin receptor (InsR)[3][7]. NVP-ADW742 has been shown in preclinical studies to enhance the sensitivity of cancer cell lines (notably small cell lung cancer and colorectal cancer) to chemotherapy and radiotherapy by inhibiting the PI3K-Akt pathway and reducing angiogenesis via suppression of VEGF expression[1][4]. The drug was developed by Novartis Pharma AG, but further clinical development was not pursued due to toxicity concerns[2]. Its primary potential indication was in neoplasms, especially small cell lung cancer[1][2].
