NVQ401 is a first-in-class bispecific antibody developed by Neuvasq Biotechnologies for the treatment of retinal vascular diseases, including diabetic macular edema (DME) and wet age-related macular degeneration (wAMD). The drug selectively activates the Wnt/β-catenin signaling cascade by targeting G protein-coupled receptor 124 (GPR124) and Low-density lipoprotein receptor-related protein 6 (LRP6), which are essential for maintaining and repairing the blood-retinal barrier. In preclinical studies, NVQ401 has demonstrated the ability to reverse VEGF-induced vascular permeability and reduce both neovascularization and avascular areas. Its potent activation of Wnt receptor signaling suggests potential for quarterly dosing, offering a potentially more durable treatment option compared to current anti-VEGF therapies.
