Overview

NVS-PI3K-3 (also referred to as NVS-PI3-3 or NVS-P13K-3) is a selective small molecule inhibitor of the delta (δ) isoform of phosphatidylinositol 3-kinase (PI3K). Developed by Novartis, it is primarily used as a research tool to investigate the role of PI3Kδ in chronic inflammatory conditions, specifically chronic obstructive pulmonary disease (COPD). In studies using alveolar macrophages and monocyte-derived macrophages from COPD patients, NVS-PI3K-3 has demonstrated the ability to significantly reduce the production of pro-inflammatory cytokines, such as TNF-α and IL-6, by inhibiting downstream AKT phosphorylation. Research indicates that this inhibition does not negatively impact the macrophages' ability to phagocytose bacteria or clear apoptotic cells, suggesting that PI3Kδ inhibitors could potentially serve as anti-inflammatory treatments that do not compromise host defense mechanisms.
