Overview

NVS-PI3K-5 is a selective small molecule inhibitor of the gamma isoform of Class I phosphatidylinositol 3-kinase (PI3Kγ). Developed by Novartis, it serves as a pharmacological tool compound used to investigate the role of PI3K signaling in inflammatory and innate immune responses. In the context of chronic obstructive pulmonary disease (COPD) research, NVS-PI3K-5 has been evaluated for its impact on alveolar macrophage functions, including the phagocytosis of bacteria (such as *Streptococcus pneumoniae* and *Haemophilus influenzae*) and the efferocytosis of apoptotic cells. Research indicates that while PI3Kγ is typically associated with leukocyte chemotaxis, its inhibition by NVS-PI3K-5 does not significantly impair the phagocytic or bactericidal capabilities of macrophages in COPD models, nor does it significantly modulate the production of pro-inflammatory cytokines like TNFα and IL-6 in those specific cell types.
