Overview

NW006-296 is a novel antibody-drug conjugate (ADC) designed to target Receptor tyrosine kinase-like orphan receptor 1 (ROR1), a protein frequently overexpressed in various hematological and solid tumors. Developed by Chengdu Chipscreen NewWay Biosciences, the agent consists of a humanized anti-ROR1 monoclonal antibody site-specifically conjugated to the microtubule-disrupting cytotoxin monomethyl auristatin E (MMAE) via a cleavable linker, maintaining a drug-to-antibody ratio (DAR) of 4. Upon binding to ROR1 on the tumor cell surface, the ADC is internalized, and the payload is released to induce cell cycle arrest and apoptosis. Preclinical studies have demonstrated that NW006-296 possesses potent anti-tumor activity and superior efficacy compared to benchmark ROR1 ADCs in models of breast cancer, colorectal cancer, and B-cell malignancies. It is currently in the IND-enabling stage of development.
