NW008 is a first-in-class dual-functional antibody-drug conjugate (ADC) targeting MHC class I polypeptide-related sequences A and B (MICA/B), which are stress-induced antigens overexpressed in various solid tumors. Developed by Chengdu Chipscreen Newway Biosciences, NW008 consists of a humanized anti-MICA/B antibody site-specifically conjugated to a potent topoisomerase I inhibitor payload via a hydrophilic, enzyme-cleavable linker. The drug features a unique dual mechanism of action: it restores antitumor immunity by inhibiting the proteolytic shedding of MICA/B from the tumor cell surface—thereby preventing immune evasion and enhancing NKG2D-mediated recognition by NK cells and T cells—while simultaneously delivering a cytotoxic payload to directly induce tumor cell death. Preclinical data have demonstrated sub-nanomolar binding affinity across multiple MICA/B alleles and significant tumor growth inhibition in xenograft models.
