NW024-1 is a novel antibody-drug conjugate (ADC) targeting placental alkaline phosphatases ALPP and ALPPL2, which are highly expressed in various solid tumors including ovarian, endometrial, gastric, and non-small cell lung cancers (NSCLC), while maintaining limited expression in normal tissues. Developed by Chengdu Chipscreen NewWay Biosciences, the agent consists of a humanized IgG1 monoclonal antibody conjugated to a topoisomerase 1 inhibitor (TOP1i) payload via a cleavable hydrophilic linker, with a drug-to-antibody ratio (DAR) of 8. NW024-1 is designed to selectively deliver the cytotoxic payload to ALPP/ALPPL2-expressing tumor cells through internalization. Preclinical studies have demonstrated potent antitumor efficacy and near-complete tumor regression in gastric cancer xenograft models, supporting its advancement into clinical development.
