NWY001 (also known as YH008) is a first-in-class bispecific antibody that targets both Programmed cell death protein 1 (PD-1) and CD40. Developed for the treatment of advanced solid tumors, it is designed to synergistically engage PD-1 and activate the CD40 pathway in a PD-1 dependent manner. This mechanism aims to reduce toxicity typically associated with CD40 agonists while potentially converting immunologically "cold" tumors into "hot" tumors, thereby enhancing patient response to PD-(L)1 immune checkpoint inhibitors—including those resistant to such therapies. The drug is currently being evaluated in Phase 1 clinical trials for various solid malignancies[1][2][4][5].
