NX-1607

NX-1607 is a first-in-class oral small molecule inhibitor of Casitas B-lineage lymphoma proto-oncogene B (CBL-B). It functions as an intramolecular glue that locks CBL-B in its closed, inactive conformation, preventing its E3 ligase activity. This inhibition results in immune activation, enhanced T cell and natural killer (NK) cell function, increased cytokine production (IL-2 and IFN-γ), reduced T cell exhaustion, and infiltration of activated immune cells in the tumor microenvironment. NX-1607 has demonstrated potent anti-tumor activity in preclinical models both as a monotherapy and in combination with anti-PD-1 therapy. Developed by Nurix Therapeutics, it is currently in clinical trials for the treatment of advanced solid malignancies.
