Drug intelligence / Profile preview
NXC03 is an artificial intelligence (AI)-designed, affinity-attenuated interleukin-21 (IL-21) mutein fused to an IgG4 Fc domain. Developed by neoX Biotech, NXC03 is designed to overcome the short half-life and high dose-limiting toxicities (such as hepatotoxicity and neutropenia) associated with wild-type IL-21. It features four amino acid mutations compared to wild-type IL-21, resulting in a unique fast-on, fast-off binding kinetics to the IL-21 receptor (IL-21R) with a 1000-fold weaker binding affinity (larger KD) and significantly improved thermal stability. In preclinical models, NXC03 demonstrated robust, dose-dependent antitumor activity both as a monotherapy and in combination with anti-PD-1 checkpoint inhibitors, while exhibiting an extended half-life and a favorable safety profile in non-human primates.
