NXD30001 is a novel, brain-penetrating small molecule inhibitor of Heat Shock Protein 90 (HSP90) developed by NexGenix Pharmaceuticals. Based on a radicicol backbone, the compound is specifically designed to cross the blood-brain barrier with improved permeability and reduced cytotoxicity compared to first-generation inhibitors like geldanamycin. NXD30001 functions by inhibiting the molecular chaperone HSP90, which subsequently leads to the ubiquitin-proteasome-mediated degradation of various client proteins essential for tumor growth and survival, such as EGFR, Akt, c-Myc, and Notch1. It has demonstrated efficacy in preclinical models of glioblastoma multiforme (GBM), both as a monotherapy and in combination with radiotherapy, and has shown neuroprotective potential in models of amyotrophic lateral sclerosis (ALS).
