NXE0027477 is a highly selective, first-in-class, oral small molecule agonist of the G protein-coupled receptor 35 (GPR35). Developed by Nxera Pharma (formerly Sosei Heptares), the compound was designed using a proprietary structure-based drug design (SBDD) platform. GPR35 is an orphan receptor with a strong genetic association to inflammatory bowel disease (IBD). Preclinical data suggest that NXE0027477 improves intestinal barrier function and reduces visceral pain, making it a potential treatment for gastrointestinal disorders such as ulcerative colitis and irritable bowel syndrome. The program was previously licensed to GSK in 2020, but full ownership was regained by Nxera in 2024. It is currently clinic-ready, having received regulatory approval from UK authorities to commence first-in-human Phase 1 studies.
