NXE0149 is a small molecule inhibitor of the NLRP3 (NACHT, LRR, and PYD domains-containing protein 3) inflammasome, developed by Nexeon MedSystems. The NLRP3 inflammasome is a multiprotein complex that plays a critical role in the innate immune system by triggering the release of pro-inflammatory cytokines, specifically interleukin-1β (IL-1β) and interleukin-18 (IL-18), and inducing pyroptosis in response to cellular stress or infection. By selectively inhibiting NLRP3, NXE0149 aims to mitigate chronic inflammation associated with various conditions, including neurodegenerative diseases like Parkinson's and Alzheimer's, as well as metabolic and autoimmune disorders. The drug is designed to cross the blood-brain barrier, making it a potential candidate for treating neuroinflammatory pathologies. It was acquired by Nexeon MedSystems in 2018 to complement their bioelectronic medicine portfolio, which focuses on neurostimulation devices.
