NxM CAR T cells are a next-generation, off-the-shelf, multi-antigen targeting chimeric antigen receptor (CAR) T-cell platform developed by Fate Therapeutics for the treatment of autoimmune diseases. Derived from a clonal induced pluripotent stem cell (iPSC) line, these cells are engineered to eliminate diverse populations of aberrant B and T cells without the need for conditioning chemotherapy. The platform incorporates multiple synthetic elements, including CARs targeting CD19 (utilizing a 1XX signaling endodomain), BCMA, and/or CD38. To enhance persistence and function in an allogeneic setting without lymphodepletion, the cells are further modified with an Allo-immune Defense Receptor (ADR), genetic disruption of the immune synapse molecule CD58 (CD58 KO), and knockout of the endogenous T-cell receptor (TCR). This approach aims to provide a scalable, outpatient-compatible 'immunological reset' for patients with systemic lupus erythematosus (SLE) and other B- and T-cell driven pathologies.
