NXP900 is an oral small molecule inhibitor targeting the SRC family of kinases (SFK), including SRC and YES1. It acts by locking SRC in its inactive "closed" conformation, thereby inhibiting both kinase-dependent catalytic activity and kinase-independent scaffolding functions. This dual inhibition distinguishes it from other multi-targeted kinase inhibitors that only block catalytic activity. NXP900 exhibits high selectivity for SFK members with sub-nanomolar potency and does not inhibit ABL kinase, which may reduce the risk of immunosuppression compared to some other agents. The drug is being developed as an anti-cancer therapy and has shown potent preclinical activity against esophageal squamous cell carcinoma (ESCC), head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC), and other advanced solid tumors[2][3][4][5].
