Overview

NY-288 is an investigational targeted alpha therapy (TAT) being developed by NAYA Therapeutics for the treatment of hepatocellular carcinoma (HCC). The therapeutic consists of a targeting agent directed against Glypican-3 (GPC3), a cell-surface oncofetal antigen that is highly overexpressed in HCC but has minimal expression in healthy adult tissues. This targeting moiety is conjugated to Astatine-211 (211At), a potent alpha-emitting radioisotope. By binding specifically to GPC3-expressing tumor cells, NY-288 delivers high-linear energy transfer (LET) alpha particles over a very short range (50-100 µm), causing complex double-strand DNA breaks that lead to cell death. This localized mechanism of action is designed to maximize anti-tumor efficacy while sparing adjacent healthy liver tissue. NY-288 is currently in the IND-enabling stage of development.
