Overview

NY-ESO-1 is a cancer-testis antigen, a protein expressed in a variety of tumors, including melanoma, synovial sarcoma, and multiple myeloma, but generally not in normal adult tissues, with the exception of testis and placenta. Its restricted expression pattern and demonstrated immunogenicity make it an attractive target for cancer immunotherapy. It is utilized in vaccine formulations to induce antigen-specific humoral and cellular immune responses, and as a target for T-cell receptor (TCR) engineered T-cell therapies, where T cells are genetically modified to recognize and attack NY-ESO-1-expressing cancer cells.
