NY-ESO-1+ CAR T cells are an investigational cell therapy in which a patient’s T cells are genetically engineered to express a chimeric antigen receptor (CAR) specifically targeting the cancer-testis antigen NY-ESO-1. NY-ESO-1 is highly expressed in many tumors (such as multiple myeloma, synovial sarcoma, neuroblastoma, and some other solid tumors) but not in most healthy tissues, making it an attractive target for immunotherapy. The CAR enables the T cells to selectively recognize and destroy NY-ESO-1 positive cancer cells. This engineered approach expands the potential for immunotherapy to target intracellular tumor antigens that are presented in the context of major histocompatibility complex (MHC), specifically HLA-A2, using a TCR-mimic CAR design. Preclinical and early-phase trials have suggested anti-tumor activity for NY-ESO-1+ CAR T cells in relevant indications, and research is ongoing in high-risk multiple myeloma and other solid tumors expressing NY-ESO-1[3][1].
