Overview

NY-ESO-1 + MPLA is a therapeutic cancer vaccine composed of the tumor antigen NY‑ESO‑1 and the adjuvant monophosphoryl lipid A (MPLA). NY‑ESO‑1 is a cancer/testis antigen highly immunogenic and expressed in various malignancies, including lung, ovarian, and melanoma. The addition of MPLA, an immunostimulatory adjuvant derived from bacterial lipopolysaccharide (LPS), enhances immune responses by activating Toll-like receptor 4 (TLR4) on antigen-presenting cells. This combination aims to induce robust cellular and humoral immunity against tumors expressing NY‑ESO‑1. Clinical trials have evaluated its safety, tolerability, immunogenicity, and preliminary efficacy in patients with cancers expressing the target antigen[2][4][8].
