NY-ESO-1 157-165 SLLMWITQA analogue is a synthetic peptide designed as an analogue of the native NY-ESO-1 157-165 peptide (SLLMWITQC). The modification involves the substitution of Cysteine (C) with Alanine (A) at the C-terminal position (position 165), resulting in the sequence SLLMWITQA. This analogue is engineered to improve its formulation stability and immunogenicity compared to the wild-type peptide. It functions as a T-cell epitope, specifically binding to the HLA-A*02:01 (HLA-A2) major histocompatibility complex (MHC) class I molecule. The HLA-A2 complex then presents the peptide to the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes (CTLs), thereby stimulating an immune response against cells expressing the cancer-testis antigen NY-ESO-1. NY-ESO-1 is a highly immunogenic tumor antigen found in various cancers, including melanoma, neuroblastoma, and other solid tumors, but with limited expression in normal adult tissues, making it an attractive target for cancer immunotherapy. This peptide analogue is utilized in the development of cancer vaccines and adoptive T-cell therapies aimed at eliciting specific anti-tumor immunity.
