Overview

NY-ESO-1 ISCOMATRIX vaccine is an investigational therapeutic cancer vaccine comprising full-length recombinant **New York esophageal squamous cell carcinoma 1** protein formulated with the proprietary saponin-based **ISCOMATRIX** adjuvant. It was developed for NY-ESO-1-expressing cancers, principally melanoma, to induce integrated NY-ESO-1-specific antibody, CD4-positive T-cell, and CD8-positive T-cell responses. Clinical studies used intramuscular administration, commonly delivering 100 micrograms of recombinant NY-ESO-1 protein with 120 micrograms of ISCOMATRIX adjuvant per injection. The vaccine was generally well tolerated and strongly immunogenic, but randomized phase 2 testing in high-risk resected melanoma did not improve relapse-free survival or overall survival versus ISCOMATRIX alone; development as a single-agent melanoma vaccine was not pursued further.
