NY-ESO-1 OLP4 is a synthetic peptide-based therapeutic cancer vaccine designed to stimulate immune responses against tumors expressing the cancer-testis antigen NY-ESO-1. The OLP4 construct consists of four overlapping 30–32 amino acid peptides that together cover residues 79–173 of the NY-ESO-1 protein, encompassing approximately 90% of known T cell epitopes and many undefined epitopes. This design allows for broad immunogenicity across diverse HLA types and aims to induce both CD8+ and CD4+ T cell responses. The vaccine has been investigated primarily in patients with epithelial ovarian cancer, fallopian tube carcinoma, and primary peritoneal carcinoma. It has been tested alone or in combination with immunoadjuvants such as Montanide (incomplete Freund’s adjuvant) and Poly ICLC (a toll-like receptor 3 agonist), as well as in combination with checkpoint inhibitors like nivolumab or ipilimumab[5][6][8]. Developed by Memorial Sloan Kettering Cancer Center, clinical development reached up to phase 2 but was ultimately discontinued[3][4][5].
