Overview

NY-ESO-1 OLP4 Vaccine + Poly-ICLC is a combination immunotherapeutic cancer vaccine composed of four overlapping long peptides (OLP4) derived from the NY-ESO-1 antigen and the immunomodulatory adjuvant polyinosinic-polycytidylic acid–poly-L-lysine carboxymethylcellulose (Poly-ICLC). The vaccine targets the cancer-testis antigen NY‑ESO‑1, which is highly immunogenic and expressed in various tumors but limited in normal tissues. The OLP4 construct covers residues 79–173 of NY‑ESO‑1, encompassing most known T-cell epitopes and avoiding HLA restriction. Poly‑ICLC acts as a Toll-like receptor 3 (TLR3) agonist to stimulate innate immunity and enhance adaptive immune responses. This combination aims to induce robust CD4+ and CD8+ T-cell responses as well as humoral immunity against tumor cells expressing NY‑ESO‑1, with primary indications in epithelial ovarian cancer, fallopian tube carcinoma, primary peritoneal carcinoma, and has also been studied in melanoma[2][4][5][7].
