Overview

NY-ESO-1 peptide is a synthetic peptide derived from the NY-ESO-1 antigen, a cancer/testis antigen that is highly immunogenic and expressed in a wide variety of tumor types but not in most adult somatic tissues. As a peptide vaccine, it is designed to elicit a specific immune response by priming T cells to recognize and kill cancer cells expressing the NY-ESO-1 antigen. NY-ESO-1 peptide vaccines have been investigated in clinical trials for various cancers, including melanoma, sarcoma, and ovarian cancer. The mechanism involves presenting the NY-ESO-1 peptide to immune cells (such as dendritic cells), which then stimulate NY-ESO-1-specific cytotoxic T lymphocytes to target and destroy tumor cells. This approach can be used as a standalone vaccine, or in combination with dendritic cell-based vaccines, checkpoint inhibitors, or engineered TCR-T cell therapies. Some vaccine constructs use long peptides (covering specific NY-ESO-1 epitopes) with or without adjuvants to enhance immune activation[1][2][7][8].
