NY-ESO-1 peptide-pulsed dendritic cells are an autologous cellular immunotherapy and cancer vaccine designed to elicit a targeted immune response against tumors expressing the New York esophageal squamous cell carcinoma 1 (NY-ESO-1) antigen. The process involves harvesting a patient's monocytes via leukapheresis, differentiating them into dendritic cells (DCs) ex vivo, and "pulsing" or loading them with specific HLA-restricted peptides derived from the NY-ESO-1 protein. NY-ESO-1 is a cancer-testis antigen that is highly expressed in various malignancies—including melanoma, synovial sarcoma, and ovarian cancer—but is absent in normal adult tissues except for the testis, making it a highly specific target. Once re-infused into the patient, these mature, antigen-presenting DCs migrate to the lymph nodes where they activate and expand NY-ESO-1-specific CD8+ cytotoxic T lymphocytes and CD4+ helper T cells to recognize and destroy tumor cells.
