NY-ESO-1 peptide vaccine is an investigational cancer immunotherapy developed by the National Cancer Institute (NCI). It targets the NY-ESO-1 antigen, a tumor-specific cancer-testis antigen expressed in various malignancies, including melanoma. The vaccine is composed of synthetic peptides, specifically the HLA-A*0201-restricted peptide (ESO-1:165V) and the HLA-DP4-restricted peptide (NY-ESO-1:161-180). These peptides are designed to stimulate CD8+ cytotoxic T-cell and CD4+ helper T-cell immune responses, respectively, against tumor cells expressing the NY-ESO-1 protein. It has been primarily evaluated in Phase II clinical trials for patients with refractory metastatic melanoma who are positive for the relevant HLA alleles.
