Overview

**NY-ESO-1 protein + GLA-SE (G305)** is a vaccine-based immunotherapy consisting of recombinant NY-ESO-1 protein, a cancer/testis antigen with high immunogenicity, combined with GLA-SE, a synthetic Toll-like receptor 4 (TLR4) agonist formulated in a stable emulsion. G305 is designed to stimulate both humoral and cellular immune responses targeting the NY-ESO-1 antigen, which is expressed in various solid tumors but mostly absent from normal tissues. The vaccine promotes the activation of CD4 T-cells, increases NY-ESO-1-specific antibody titers, and aims to induce anti-tumor immunity. G305 has demonstrated safety and immunogenicity in Phase 1 clinical trials with patients exhibiting NY-ESO-1-expressing tumors, showing potential clinical benefits such as disease stabilization[1][3][10].
