NY-ESO-1 reactive TCR retroviral vector transduced autologous PBL is an investigational **autologous T-cell receptor engineered T-cell therapy** developed at the Jonsson Comprehensive Cancer Center. A patient's peripheral blood lymphocytes are genetically modified with a retroviral vector to express an NY-ESO-1-reactive T-cell receptor, then reinfused after lymphodepleting conditioning. The engineered T cells are intended to recognize NY-ESO-1-positive tumor cells through antigen presentation by HLA class I and mediate antigen-specific cytotoxic antitumor activity. The product has been studied with separately administered NY-ESO-1 peptide-pulsed dendritic-cell vaccination and low-dose interleukin-2 in patients with advanced NY-ESO-1-expressing malignancies.
