Overview

NY-ESO-1 reactive TCR retroviral vector transduced autologous T cells is an investigational adoptive cell transfer therapy developed by the Jonsson Comprehensive Cancer Center at UCLA. The therapy involves harvesting a patient's own peripheral blood lymphocytes (PBLs), which are then genetically engineered using a retroviral vector to express a specific T-cell receptor (TCR) designed to recognize the NY-ESO-1 antigen. NY-ESO-1 is a cancer-testis antigen that is highly expressed in various solid tumors but has restricted expression in normal tissues, making it an ideal target for immunotherapy. Once these modified T cells are expanded and re-infused into the patient, they target and eliminate cancer cells presenting NY-ESO-1 peptides in the context of HLA-A*0201. In clinical settings, this therapy is often evaluated in combination with other agents, such as the CTLA-4 inhibitor ipilimumab and dendritic cell vaccines, to overcome immunosuppression and enhance the persistence and activity of the engineered T cells within the tumor microenvironment.
