Overview

This investigational autologous adoptive T-cell therapy consists of patient-derived CD4+ T cells that are engineered or selected to specifically recognize the NY-ESO-1 (New York esophageal squamous cell carcinoma 1) cancer-testis antigen. Developed by the M.D. Anderson Cancer Center, the therapy targets NY-ESO-1-expressing tumors, including melanoma, synovial sarcoma, and myxoid/round cell liposarcoma. The mechanism involves the infusion of these antigen-specific CD4+ T cells, which can mediate direct tumor regression and orchestrate a broader immune response against multiple tumor antigens. In clinical studies, it is evaluated following lymphodepletion with cyclophosphamide and in combination with the checkpoint inhibitor ipilimumab to enhance T-cell persistence and anti-tumor activity.
