NY-ESO-1 T-cell receptor mimic antibody is a fully human monoclonal antibody developed by Biocytogen using its proprietary RenMab mouse platform and TCR-mimic (TCRm) technology. The antibody is engineered to recognize the intracellular cancer-testis antigen NY-ESO-1 (specifically the 157-165 peptide SLLMWITQC) when presented on the cell surface by the HLA-A*02 MHC class I molecule. By mimicking the specificity of a T-cell receptor but with the high affinity characteristic of a monoclonal antibody (nanomolar range), it overcomes the low-affinity limitations of natural TCRs. This candidate is primarily intended for use in cell therapy, such as TCRm-engineered T cells (TCRm-T), to target and eliminate NY-ESO-1-expressing tumor cells in HLA-A02-positive patients. It has demonstrated potent and specific anti-tumor activity in preclinical models of synovial sarcoma, melanoma, and neuroblastoma.
