Overview

NY-ESO-1 targeted T cells refer to an autologous adoptive T-cell therapy designed to treat cancers expressing the New York Esophageal Squamous Cell Carcinoma 1 (NY-ESO-1) antigen. This specific program, investigated by The Christie NHS Foundation Trust, involves the isolation of a patient's own T cells, which are then engineered or selected to recognize the NY-ESO-1 cancer-testis antigen in the context of HLA-A2. NY-ESO-1 is a highly immunogenic protein with expression typically restricted to germ cells but frequently overexpressed in various malignancies, including oesophagogastric cancer. The therapy was evaluated in a Phase II clinical trial (ATTACK-OG; NCT01795976) for patients with advanced gastroesophageal cancer, but the study was terminated in 2017 due to a withdrawal of funding.
