Overview

NY-ESO-1 TCR + IL-15 NK is an investigational, off-the-shelf cell therapy composed of cord blood-derived natural killer (NK) cells that are genetically engineered to express an affinity-enhanced T-cell receptor (TCR) targeting the cancer-testis antigen NY‑ESO‑1 and to secrete interleukin 15 (IL‑15). The therapy is designed to combine the innate tumor-targeting ability of NK cells with the antigen specificity of a TCR directed at NY‑ESO‑1, which is highly expressed in several cancers but has limited expression in normal tissues. The addition of IL‑15 supports enhanced survival and proliferation of the engineered cells. This approach aims to improve anti-tumor activity while reducing risks associated with conventional CAR-T or autologous therapies. It is being developed for relapsed or refractory multiple myeloma as well as advanced synovial sarcoma and myxoid/round cell liposarcoma[4][5][6][9].
