Overview

NY-ESO-1 TCR engineered cells are a type of autologous cell therapy in which a patient’s T cells are isolated, genetically engineered to express a T-cell receptor (TCR) specific for the cancer-testis antigen NY-ESO-1, and then reinfused into the patient. The engineered TCR enables the T cells to selectively recognize and bind to NY-ESO-1 peptides presented by HLA-A*02:01 (or related alleles) on the surface of tumor cells, leading to targeted tumor cell killing. Primary indications include synovial sarcoma, melanoma, multiple myeloma, and other NY-ESO-1-expressing solid tumors. Early phase trials and clinical reports have demonstrated objective and durable clinical responses, especially in synovial sarcoma and melanoma. The approach is distinct from CAR-T therapy as it targets intracellular peptides presented by HLA molecules rather than surface antigens. Multiple research organizations and companies have developed versions of NY-ESO-1-specific TCR products, employing affinity-enhancement, gene engineering (for example, retroviral transduction), and silencing of endogenous TCRs to improve safety and efficacy.
