**NY-ESO-1 TCR-engineered peripheral blood mononuclear cells** are an investigational autologous T-cell therapy developed by the Jonsson Comprehensive Cancer Center at UCLA for NY-ESO-1-positive advanced malignancies. Patient-derived peripheral blood mononuclear cells are genetically modified to express a T-cell receptor recognizing the cancer-testis antigen NY-ESO-1, enabling redirected T-cell recognition and killing of NY-ESO-1-expressing tumor cells in an HLA-restricted context. In the UCLA NYESO SCT Phase I protocol, this PBMC product is administered intravenously after myeloablative conditioning as part of a tandem cellular-treatment approach with separately engineered autologous CD34-positive peripheral blood stem cells and interleukin-2 support. This UCLA investigational product is distinct from Takara Bio's mipetresgene autoleucel.
