NY-ESO-1 TCR-engineered T-cell therapy is an investigational adoptive cell therapy developed by the Jonsson Comprehensive Cancer Center (UCLA) for the treatment of advanced malignancies expressing the NY-ESO-1 antigen. The therapy utilizes autologous peripheral blood lymphocytes (PBL) that are genetically modified via a retroviral vector to express a T-cell receptor (TCR) specific for the NY-ESO-1:157-165 peptide presented by HLA-A*0201. In clinical protocols (such as NCT00924326), this cell therapy is administered following a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine. The treatment is further augmented by the administration of a dendritic cell vaccine pulsed with the NY-ESO-1 peptide and systemic aldesleukin (IL-2) to promote the expansion and persistence of the engineered T cells in vivo.
