Overview

NY-ESO-1 TCR T-cell therapy is an adoptive cell immunotherapy involving the genetic modification of a patient's T cells to express a T-cell receptor (TCR) specific for the cancer-testis antigen NY-ESO-1 (New York esophageal squamous cell carcinoma 1). NY-ESO-1 is a tumor-associated antigen that is highly expressed in various cancers but has restricted expression in normal tissues, making it an ideal target for immunotherapy. The therapy typically utilizes a viral vector (retroviral or lentiviral) to introduce an affinity-enhanced TCR that recognizes NY-ESO-1 peptides presented by HLA-A*0201 molecules. Clinical development has primarily focused on synovial sarcoma, myxoid/round cell liposarcoma (MRCLS), and multiple myeloma, where it has demonstrated significant objective response rates. Major programs include Adaptimmune's letetresgene autoleucel (formerly partnered with GSK) and Takara Bio's mipetresgene autoleucel (TBI-1301).
