NY-ESO-1 TCR-T cells are a form of adoptive cell therapy that uses genetically modified T cells expressing a T-cell receptor (TCR) specifically targeting the NY-ESO-1 antigen. NY-ESO-1 is a cancer-testis antigen that is not expressed in normal, non-germline tissue but is aberrantly expressed in many tumors, particularly in soft tissue sarcomas (especially synovial sarcoma), melanoma, and multiple myeloma. The therapy involves collecting a patient's own T cells, genetically modifying them to express a TCR that recognizes the NY-ESO-1 antigen presented by specific HLA types (commonly HLA-A*02:01 or HLA-A*02:06), expanding these cells in the laboratory, and then reinfusing them into the patient after lymphodepletion chemotherapy. Some versions of the therapy include affinity-enhanced TCRs and/or silencing of endogenous TCRs to improve efficacy and safety.
