This investigational cancer vaccine, developed by Dr. Hassane M. Zarour at the University of Pittsburgh, targets the NY-ESO-1 (New York esophageal squamous cell carcinoma-1) antigen, a cancer-testis antigen frequently expressed in various solid and hematologic malignancies. The vaccine formulation consists of either NY-ESO-1-derived peptides or recombinant NY-ESO-1 protein, combined with two adjuvants: CpG 7909 (PF-3512676), a Toll-like receptor 9 (TLR9) agonist, and Montanide ISA 720, a water-in-oil emulsion. In some clinical protocols, low-dose cyclophosphamide is administered as an immunomodulator to enhance the vaccine's efficacy by depleting regulatory T cells. The vaccine is designed to stimulate robust tumor-specific immune responses, including CD4+ and CD8+ T cell activation and antibody production, to target and eliminate NY-ESO-1-expressing cancer cells.
